Itchiness explained: Specific set of nerve cells signal itch but not pain, researchers find

Jan. 2, 2013 ? Johns Hopkins researchers have uncovered strong evidence that mice have a specific set of nerve cells that signal itch but not pain, a finding that may settle a decades-long debate about these sensations, and, if confirmed in humans, help in developing treatments for chronic itch, including itch caused by life-saving medications.

At the heart of their discovery is a type of sensory nerve cell whose endings receive information from the skin and relay it to other nerves in the spinal cord, which then coordinates a response to the stimulus. Published online Dec. 23 in Nature Neuroscience, a report on the research suggests that even when the itch-specific nerve cells receive stimuli that are normally pain-inducing, the message they send isn?t ?That hurts!? but rather ?That itches!?

Pain and itch are both important sensations that help organisms survive. And pain is arguably more important because it tells us to withdraw the pained body part in order to prevent tissue damage. But itch also warns us of the presence of irritants, as in an allergic reaction. However, ?when either of these sensations continues for weeks or months, they are no longer helpful. We even see patients stop taking life-saving medications because they cause such horrible itchiness all over,? says Xinzhong Dong, a Howard Hughes early career scientist and associate professor of neuroscience at the Institute of Basic Biomedical Sciences at the Johns Hopkins University School of Medicine. ?And sometimes when we try to suppress chronic pain, with morphine for example, we end up causing chronic itchiness. So the two sensations are somehow related, and this study has begun to untangle them,? he says.

Because nerve cells send their messages as electrical currents that flow through them just as they would through wires, scientists can plug tiny monitors into individual nerve cells to detect the moment of stimulation. The scientific controversy over pain and itch centers around a group of nerve cells known to respond electrically to painful stimuli such as molecules of capsaicin, the fiery ingredient in chili peppers. A small subset of these nerve cells also responds electrically to itchy stimuli because they have on their surfaces receptors for molecules like histamine. One of these itchy receptors, called MrgA3, binds the anti-malaria drug chloroquine, causing serious itchiness in many patients.

Sensory nerve scientists have not known whether the nerves with itchy receptors and pain receptors were actually sending both types of messages to the brain, or just itch messages. What the current study found is that, in nerves with the itchy receptor MrgA3, electrical signals sent in response to both painful and itchy stimuli are interpreted by the brain as itch.

To reach this conclusion, the researchers first used a genetic trick to label the MrgA3 cells in mice with a glowing protein that allowed them to see the cells under the microscope. Aided by the glow, they were able to plug in those tiny electricity monitors and watch nerve cell responses to different stimuli. The cells transmitted electrical signals when the mice were exposed to itch-inducing chloroquine and histamine, as well as pain-inducing capsaicin and heat. Based on this result, the researchers tentatively concluded that the cells could send both pain and itch signals.

In the next experiment, the researchers monitored the behavioral responses of mice to the different stimuli. As expected, when the tails of normal mice were placed in hot water, they quickly pulled them out; when normal mice were given a bit of chloroquine or histamine, they scratched vigorously with their hind legs.

Then, to examine the role of MrgA3 cells in pain and itch, the scientists selectively killed MrgA3 nerve cells in adult mice and retested their responses. Presumably, the researchers noted, because MrgA3 cells are only a small fraction of all pain-sensing nerve cells, the mice had normal withdrawal responses to painful stimuli like hot water. However, when exposed to itchy stimuli, their scratching responses were reduced to varying degrees depending on the stimulus, most significantly in response to chloroquine. The fact that some stimuli still caused scratching suggested to the scientists that MrgA3 cells are not the only ones in the body that respond to itch. ?We were convinced that MrgA3 cells are responsible for much of the sensation of itch, but it wasn?t yet clear whether MrgA3 cells could also relay painful information,? says Dong.

In their final experiments, the scientists used genetic techniques to create mice in which the MrgA3 cells were the only cells in the body capable of responding to capsaicin, that peppery pain-inducing substance. When injected into the cheeks of mice, normal mice massage the area with their forepaws to relieve the hot sensation. When injected into the experimental mice, they vigorously scratched their cheeks with their hind legs, suggesting that this normally painful stimulus had been communicated to the brain?by MrgA3 cells?as itchiness.

"Now that we have disentangled these itchy sensations from painful ones, we should be able to design drugs that target itch-specific nerve cells to combat chronic itchiness," says Dong. "We hope that this will not only provide relief, but also increase people's faithfulness to their drug plans, particularly for deadly diseases like malaria and cancer."

3 bright spots in the US job market for 2013 - Fortune Finance ...

The unemployment rate peaked at 10% in late 2009. Since then, it has only fallen to 7.7% as of November, suggesting the long slog is likely to continue into the New Year.

For 2013, economists widely agree the outlook for jobs will continue improving but not by much. The economy is expected to create 2.36 million jobs, slightly more than the estimated 1.9 million that's estimated for 2012, according to IHS Global Insight.

Older workers, particularly those who've been out of work for more than six months, have been disproportionately impacted. So have young workers struggling to launch their careers. And countless other groups are feeling the weight of an economy peeling off from a financial crisis and housing market meltdown.

Amid another blah year, a few workers that bore the brunt of the Great Recession stand to gain in 2013. Here are three:

College graduates

Congrats class of 2013! You have a better chance landing your dream job than the poor grads before you working as baristas.

The years following the Great Recession hit workers across the board, but those stepping out of college campuses in 2012 were hit particularly hard. They graduated at a time when half of all recent grads were either jobless or working low-waged jobs, according to an April analysis by the Associated Press.

The job market isn't expected to be quite as harsh for young people next year, economists say. Employers anticipate hiring 13% more Class of 2013 college graduates than they hired from the Class of 2012, according to a survey of employers conducted by the National Association of Colleges and Employers.

The top five bachelor's degree in demand? Finance, followed by computer and information science, accounting, business administration/management and mechanical engineering.

Construction workers

As the housing market continues to heal in 2013, expect more hiring in construction. Since the Great Recession technically ended in June 2009, jobs in homebuilding dropped 14.5% from 633,000 to 552,800 in November, according to the Labor Department.

In 2012 alone, the construction industry lost 9,000 jobs. That's expected to reverse come 2013, when the economy will likely add 100,000 jobs in homebuilding and other construction work, says Marisa Di Natale, economist at Moody's Analytics. The most dramatic pick-up is expected in 2014, with 300,000 more jobs.

Government workers

For the past three years, local and state governments held off on new hires and shed jobs almost every month. Officials struggled with plummeting tax revenues linked closely to property values.

But after years of virtually no hiring and as the housing market recovers, governments are expected to hire again, Natale says. For 2013, state governments, in particular, are expected to add 70,000 jobs.

Previous studies on toxic effects of BPA couldn't be reproduced

Jan. 2, 2013 ? Following a three-year study using more than 2,800 mice, a University of Missouri researcher was not able to replicate a series of previous studies by another research group investigating the controversial chemical BPA. The MU study is not claiming that BPA is safe, but that the previous series of studies are not reproducible. The MU study, published in the Proceedings of the National Academy of Sciences, also investigated an estrogenic compound found in plants, genistein, in the same three-year study.

"Our findings don't say anything about the positive or negative effects of BPA or genistein," said Cheryl Rosenfeld, associate professor of biomedical sciences in MU's Bond Life Science Center. "Rather, our series of experiments did not detect the same findings as reported by another group on the potential developmental effects of BPA and genistein when exposure of young occurs in the womb."

Creating reliable data on the effects of the chemicals on mice is important to human health since people are frequently exposed to BPA and genistein and humans share similar biological functions with mice. BPA is a chemical used in certain plastic bottles and may be found in the lining of some canned goods and receipt paper. Genistein occurs naturally in soy beans and is sold as a dietary supplement. Research by Fredrick VomSaal, professor of biological science at MU, and others suggests the chemicals may have other adverse effects on many animals, including humans.

Researcher who conducted the original series of experiments claimed that exposure to BPA and genestein resulted in yellow coat color, or agouti, offspring that were more susceptible to obesity and type 2 diabetes compared to their brown coat color, healthy siblings. However, Rosenfeld and her team did not obtain the same results when repeating the study over a three-year period.

After failing to repeat the original experiments findings with similar numbers of animals, Rosenfeld's group extended the studies to include animal numbers that surpassed the prior studies to verify that their findings were not a fluke and to provide sufficient number of animals to ensure that significant differences would be detected if they existed. However, even these additional numbers of animals and extended experiments failed to reproduce the earlier findings. However, the current studies demonstrate that a maternal diet enriched in estrogenic compounds leads to a greater number of offspring that express an agouti gene compared to those that do not, even though equal ratios should have been born.

"This finding suggests that certain uterine environments may favor animals with a 'thrifty genotype' meaning that the agouti gene of mice may help them survive in unfavorable uterine environments over those mice devoid of this gene, Yet, the downside of this expression of the agouti during early development is that the animals may be at risk for later metabolic disorders, such as obesity and diabetes" Rosenfeld said. "In this aspect, humans also have an agouti gene that encodes for the agouti signaling protein (ASIP) that is expressed in fat tissue and pancreas, and there is some correlation that obese individuals exhibit greater expression of this gene compared to leaner individuals. Therefore, the agouti gene may have evolved to permit humans the ability to survive famine, but its enhanced expression may also potentiate metabolic diseases under bountiful food conditions."

While the research casts doubt on the previous study, Rosenfeld said that by understanding the genetic profile of the mice in the first series of studies, scientists could learn more about the correlation between certain genes and obesity. This could eventually influence prevention and treatment programs for patients with diabetes and other obesity-related diseases in humans.

Criminal injustice: Idle No More, the prison system and Indigenous ...

Idle No More is forcing many Canadians to be Willfully Blind No More.?

Ostensibly, the movement spearheaded by Attawapiskat Chief Theresa Spence is about the protection of First Nations' treaty rights. At its core, though, might be something more profound, perhaps best described as a demand for all of us in this country to re-think the relationship between Aboriginal and non-Aboriginal Canadians. The relationship is, of course, an uneven one, though it might be far more uneven than most of us in this country care to acknowledge.?

Against this backdrop, policy makers and average Canadians alike would be well advised to ring in the New Year by considering the lessons Michelle Alexander highlights in her important book, The New Jim Crow: Mass Incarceration in the Age of Colorblindedness. The context is American, but the lessons may hit closer to home.?In the book, Alexander writes the following in regards to African-Americans and the United States' criminal justice system:?

"It is fair to say that we have witnessed an evolution in the United States from a racial caste system based entirely on exploitation (slavery), to one based largely on subordination (Jim Crow), to one defined by marginalization (mass incarceration). While marginalization may sound far preferable to exploitation, it may prove to be even more dangerous. Extreme marginalization, as we have seen throughout world history, poses the risk of extermination."

It's a narrative equally provocative and compelling. Alexander argues that Americans' obsession with 'law and order' began as a conservative attempt to breed white opposition to the burgeoning civil rights movement. Politicians on the right labeled civil disobedience a precursor to criminality, and branded supporters of civil rights legislation as promoters of lawlessness. Exploiting the fears of poor and rural whites, Republican politicians invented and propagated the 'War on Crime' and, more specifically, the 'War on Drugs' as a means to incarcerate, and thus control, the black population.

The scheme was so successful that any politician that questioned the effectiveness or morality behind the supposedly colourblind 'War on Drugs' was accused of being 'soft on crime' -- the death knell for one seeking public office. In 1992, Bill Clinton recognized that Democrats could peel off right leaning white voters by outflanking the Republicans on issues like crime and welfare reform, and promptly advocated for a number of policies that resulted in the largest increases in federal and state prison inmates of any president in the history of the United States, the overwhelming majority of which were people of colour. The normalization of racial marginalization, according to Alexander, was finally, quietly complete.?

The book is more than a lament; it's a warning. Alexander paints a bleak portrait of a future in which the majority of African Americans -- increasingly economically vulnerable in a nation of skyrocketing inequality -- being under the control of the criminal justice system is not entirely inconceivable. It's an ugly commentary on contemporary American society. Canadians should pay attention.?

Troublingly, there are significant parallels between the plight of African Americans in the United States, and the Aboriginal population in Canada.?

As American history is steeped in institutional savagery toward African Americans, Canada?s history, too, is pockmarked by the white majority's mistreatment of Indigenous peoples. Most notable among a litany of gross violations of justice are the permanent occupation of ancestral lands beginning in the eighteenth century, the noxious attempt to 'civilize' Aboriginal people in residential schools in the early 1900s, and the refusal to recognize Aboriginal Canadians as legal 'persons' until the middle of the twentieth century. And the similarities, unfortunately, are not only historical.?

Though Canada doesn't yet lock up the same percentage of its citizenry as its southern neighbor --the most heavily incarcerated society in the world -- measures passed by Prime Minister Harper's Conservative government have resulted in a steady increase in the number of Canadians behind bars. Canadians of Aboriginal ancestries have borne the brunt of the surge of prisoners.?

The proportion of African Americans and Aboriginal Canadians in prison far exceed their representation in the U.S. and Canada, respectively. In the U.S., black Americans account for 12 per cent of the total population, but represent nearly 40 per cent of the prison population. In Canada, Aboriginal peoples make up four percent of the total population, but 20 per cent of the prison population.?

These significant discrepancies are not shrinking. In the U.S., over 40 per cent of new offenders to state and federal prisons are African American. In Canada, more than one in five new admissions to men's prisons are of Aboriginal descent. Among women offenders, over one in three are Aboriginal. In the U.S., one in three black men can expect to be incarcerated at some point in their lives. In Canada, young Aboriginal men are more likely to go to prison than complete a high school diploma. And once in custody, a significantly larger proportion of African American and Aboriginal Canadian prisoners are recommended for maximum levels of security than white prisoners charged with similar crimes.?

While it might be unfair to claim that Canadian politicians and the Canadian criminal justice system have joined forces to consciously ensure that our Aboriginal communities remain repressed and disenfranchised, it cannot be denied that Aboriginals across the country suffer from poverty, injustice and lack of opportunity more than other type of Canadian. One in four Aboriginal children live in poverty.

The life expectancy for Indigenous people in Canada is eight to twenty years less than non-Aboriginal Canadians. On First Nations reserves, sixty percent of children do not graduate high school, and the lack of suitable housing and access to clean water is endemic. To be Aboriginal in Canada?today is to be a second-class citizen.?

As a new year begins, we should take this opportunity to look closely at the society that we have created, and address the appalling inequalities of our criminal justice system. There is still time to prevent the prison state of our southern neighbors, but only if we hurry.?

No Canadian should be idle.?
